For sequencing long DNA, even an entire chromosome. Not reliable in case of DNA with lots of repetitions (need to use the sequencing method based on BACs). Steps Random DNA fragmenation (restriction enzymes or physical fragmenation) Sequencing of individual fragments. Assembly of the fragments sequences by computer on the basis of their overlapping regions.
