For sequencing long DNA, even an entire chromosome.
Not reliable in case of DNA with lots of repetitions (need to use the sequencing method based on BACs).
Steps
- Random DNA fragmenation (restriction enzymes or physical fragmenation)
- Sequencing of individual fragments.
- Assembly of the fragments sequences by computer on the basis of their overlapping regions.
